It is possible to perform perforated patch clamp recordings on human-induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) with success rates up to 40%.

  • We can distinguish cells that are in perforated patch from cells in standard whole-cell (WC) using an intracellular blocker.
  • Voltage-gated Ca2+ currents are larger in perforated patch compared to standard WC.
  • Action potential measurements present longer action potential duration (APD) in perforated patch compared to standard WC, likely due to the increased contribution of L-type calcium channels
