QPatch II can be used to characterize hiPSC-derived excitatory neurons (iCell® Induced Excitatory Neurons) and uncover disease phenotypes, including time-dependent variations in ligand-gated and voltage-gated ion channel currents and action potential parameters. The results include:
- A QPatch II assay for evaluating disease phenotypes in hiPSC neurons yielding a whole-cell success rate of ~ 60 %
- An ion-channel analysis of voltage-gated (KV and NaV ) and ligand-gated (AMPA) ion channel currents over time in culture
- A proof-of-concept analysis of neuron excitability based on single-cell action potential measurements
